阿尔茨海默病(Alzheimer’s disease, AD)是最常见的进行性神经退行性疾病,占成人痴呆症的60%以上。随着人口老龄化,AD患病人数急剧增加,不但严重降低了老年人的生活质量,而且给社会和家庭带来沉重的负担。因此,寻求有效的防治措施具有重要的医学价值和社会意义。
先于神经元死亡的进行性轴突退化是AD等神经退行性疾病中普遍存在的主要的病理学事件。轴突的正常功能为从胞体向轴突远端和同时也从轴突远端向胞体运输各种功能性分子以及通过形成突触从而在神经元之间或神经元与效应细胞之间传递信息,以实现神经元多种至关重要的功能。轴突发生是指神经发育及神经再生中轴突的生成和生长,为神经元保持极性结构的关键。因此,寻找轴突发生的关键分子并阐明其作用机制对AD等神经退行性疾病中神经元轴突的保护和轴突退化的防治有着重要的意义。
